尼美舒利对人舌鳞状细胞癌Tca8113细胞ang-2基因表达的影响

Influence of Nimesulide on expression of ang-2 gene in human tongue squamous cell carcinoma Tca8113

目的探讨环加氧酶(COX)-2抑制剂尼美舒利(NIM)对人舌鳞状细胞癌Tca8113细胞中ang-2基因表达的影响及其意义。方法以25、50、100和200μmol.L-1浓度的NIM处理Tca8113细胞48 h,半定量RT-PCR检测其ang-2 mRNA的表达。结果 Tca8113细胞中ang-2 mRNA水平下降,且下降呈浓度依赖性。结论 NIM下调ang-2基因表达以抑制肿瘤血管发生,可能是COX-2抑制剂抑瘤的作用机制之一。

Objective To investigate the influence of Nimesulide（NIM）,a selective cyclo-oxygenase（COX）-2 inhibitor,on angiogenins（ang） gene expressions in human tongue squamous cell carcinoma Tca8113.Methods Tca8113 cells were treated with NIM（25,50,100,200 μmol·L-1） for 48 h to meaure the expression of ang-2 mRNA in Tca8113 cells by the semiquantitative reverse transcription polymerase chain reaction analysis.Results The expression of ang-2 mRNA reduced in a concentration dependent manner.Conclusion NIM can down-regulate ang-2 gene expression,which may be involved in cancer angiogenesis inhibition mechanisms of COX-2 inhibitor.
