摘要
目的:了解我院临床分离的铜绿假单胞菌的耐药性及其所产β-内酰胺酶的基因型。方法:采用VITEK32型全自动细菌鉴定仪对临床分离的26株铜绿假单胞菌进行细菌鉴定,采用琼脂扩散法检测其对16种常用抗菌药物的耐药性,采用聚合酶链反应法对其进行基因检测,靶基因为TEM、SHV、OXA、PER、VEB、GES、CTX-M-1等7种β-内酰胺酶基因,然后对检测的阳性产物进行测序,并对照GenBankDNA序列数据库确定基因类型。结果:26株细菌均鉴定为铜绿假单胞菌,耐药性分析结果表明其对其中12种常用抗菌药物有不同程度的耐药性,通过基因对照确定26株铜绿假单胞菌所产的β-内酰胺酶均为TEM-1型基因。结论:本组26株铜绿假单胞菌所产β-内酰胺酶可能主要为TEM-1型基因,具有多重耐药性。
OBJECTIVE: To investigate the drug resistance of clinical isolated Pseudomonas aeruginosa and the genotypes of β-lactamases produced by them in our hospital. METHODS : 26 stains of clinical isolated Pseudomonas aeruginosa were identified with VITEK 32 automated bacteria identification instrument. The Kirby-Bauer method was used to detect susceptibility of 16 kinds of antimicrobial. And polymerase chain reaction (PCR) analysis was used to detect 7 kinds of β-lactamases genes including TEM, SHV, OXA, PER, VEB, GES, CTX-M-1. DNA sequencing was used to detect positive production of PCR with GenBank as control. RESULTS : Results of genotype analysis showed that 26 strains of Pseudomonas aeruginosa carried genes of TEM-1, and isolated Pseudomonas aeruginosa were resistant to 12 kinds of common antimicrobial drugs. CONCLUSION: Genotype of β-lactamases produced by Pseudomonas aeruginosa is mainly TEM-1 with multidrug resistance.
