摘要
目的:考察罗哌卡因/乳酸羟基乙酸共聚物微球(RVP/PLGA-MS)在小鼠体内的生物相容性。方法:取昆明小鼠40只,随机均分为A、B2组,分别于小鼠一侧下肢手术切开暴露坐骨神经后注入RVP溶液和植入RVP/PLGA-MS(以RVP计每只小鼠给药量约为0.48~0.5mg)后缝合。观察小鼠手术切口的红肿、渗出情况以及愈合时间;1周后比较2组小鼠体重;2周后处死小鼠,电镜下观察2组小鼠局部组织切片及B组全身主要脏器的病理学改变。结果:2组小鼠手术切口愈合情况及体重比较无统计学差异,2组局部组织切片未见明显的病理学改变;B组全身主要脏器均无病理学改变。结论:RVP/PLGA-MS在小鼠体内具有良好的组织相容性。
OBJECTIVE: To study the biocompatibility of Ropivacaine/poly (lactide-co-glycolide) microspheres (RVP/PLGA- MS) in mice in vivo. METHODS: 40 Kunming mice were randomly divide into group A and B. In group A, the sciatic nerve exposed by surgical incision, and sutured after injecting RVP; in group B, the sciatic nerve exposed by surgical incision, and implanted with RVP/PLGA-MS, then sutured (0.48 to 0.5 mg of RVP). The inflammatory reaction, exudation and healing time of operative incision were observed. One week later, body weight of mice was compared between 2 groups. Mice were sacrificed after 2 weeks. The pathological changes of major organs in group B and local tissue slice of 2 groups were observed under light microscope. RESULTS: There was no statistical difference in incision healing and body weight between 2 groups. No significant pathological changes were found in local tissue slices of 2 groups. There were no pathological changes of major organs in group B. CON- CLUSION: RVP/PLGA-MS in mice has a good biocompatibility in vivo.
