摘要
枯萎病是甜瓜等瓜类作物的毁灭性病害,Fom-2基因介导了甜瓜抗枯萎病生理小种0和1,抗枯萎病育种是甜瓜等瓜类作物育种的重要目标之一。通过同源克隆方法从薄皮甜瓜白啄瓜中克隆到了枯萎病抗性基因Fom-2,推定编码1 099个氨基酸,所推定的蛋白质序列经Blast分析与数据库中NBS-LRR类R基因具有较高的同源性。进一步序列分析FOM-2蛋白具有典型的R基因的NB-ARC及SCOP 2个结构功能域。实时荧光定量PCR结果表明,白啄瓜Fom-2基因经枯萎病接种后表达明显上调。白啄瓜Fom-2基因为白啄瓜抗病育种奠定了基础。
Fusarium wilt (Fusarium oxysporum f. sp. melonis Snyder and Hansen) is one of destructive disease in melon crop production. Fore-2 gene was genetically identified to control the resistance of races 0 and 1. Resistance to fusarium wilt breeding is one of important targets for melon breeding. In this study, we cloned the Fore-2 gene from Baizhuogua melon (Cucumis melo ssp. agrestis Jeffrey) by using homo- logical cloning method, which was deduced to encode 1099 amino acids and exhibited high similarity to NBS-LRR type of R genes in plant. The deduced FOM-2 protein contained NB-ARC and SCOP typical motifs of R gene. The expression of Fore-2 gene was highly induced after inoculation with fore race 1. In eonclusion, the cloning of Fore-2 gene is good for breeding with resistance to fusarium wilt.
