大鼠慢性肾衰竭模型的建立及体会 被引量:12

摘要 目的观察应用大鼠制备慢性肾衰竭模型的效果。方法采用5/6肾切除方法制备慢性肾衰竭模型,分别于术后3周和8周后观察肾功能以及肾脏病理变化。结果造模成功率均达83%以上。手术组3周后出现肌酐(Scr)、血尿素氮(BUN)升高,8周后血清钙降低,血清磷、肌酐(Scr)、血尿素氮(BUN)水平明显升高,与对照组比较均有统计学意义;手术组镜下出现典型肾小球、肾小管-间质损伤的表现,系膜/肾小球面积比值、肾小管-间质损伤指数与对照组比较有统计学意义。结论应用5/6肾切除可制备稳定的慢性肾衰竭动物模型。 Objective To develop rat model of chronic renal failure, and observe the effect of the model. Methods Chronic renal failure rats model were induced by 5/6 nephrectomy, two groups of animals were observed at 3week and 8week respectively after operation, renal function and pathology of the rats were examined, Results The rate of successful model was reached 83% in model constructing experiment. Levels of serium creatini^ae (Scr) and blood urea nitrogen (BUN) were remarkablely in- creased in model group compared with thatof sham - operated control group, no matter in 3week or 8week (P 〈0.01 ). Kidneys from m0del group presented typical appearance of glomerular and tubular - interstitial injury by naked eye exomination. The rate of mesangium area and glomerular area and the index of histology score for tubular interstitial damage in model group was much higher than that of sham - operation control group (P 〈 0.01 ). Conclusions Stable rat model of chronic renal failure can be developed by 5/6NX.
