禽Ⅰ群腺病毒血清1型株灭活疫苗的研制 被引量:4

摘要 优化病毒的增殖条件,获得108.14 TCID50/mL,病毒粒子数为108.74 copies/mL的FAV-1抗原液,制备禽I群腺病毒血清1型(FAV-1)油乳剂灭活苗。比较甲醛及β-丙内酯的灭活效果,并进行疫苗成分最佳配比的优化,最后确认HLB 7.0,乳化剂含量10%,油水比为4∶1的比例稳定性最好、免疫效果最佳。疫苗免疫抗体水平的监测结果表明接种后第4周抗体水平达到最高峰,并在接种后第8周时仍具有较高抗体水平。攻毒试验结果显示,该疫苗对免疫组的保护率为100%,能完全抵抗病毒攻击。田间试验表明该疫苗达到预期免疫抗体水平,在接种后4个月保持高效价,到6个月时保持中等水平。本研究所研制疫苗免疫效果好,保存期长,安全性高。 An inactivated oil-emulsion vaccine was developed from fowl adenovirus group I serum type 1(FAV-1).After optimization of virus replication condition,the virus with the titer of 10^8.14 TCID50/mL and concentration of 108.74 copies/mL was used to prepare the vaccine.The developed vaccine with hydrophile-lipophile balance number(HLB) 7.0,10% emulsifier and oil-water ratio of 4∶1 can provide stable immune protection.The monitoring results of antibody level showed that the antibody level of immunized chicken group reached the peak value after 4 weeks.After 8 weeks,the antibodies still kept in high level.The vaccination and challenge test confirmed that the FAV-1 vaccine could protecte fowl from the attack of FAV-1 completely.The results of field experiments showed that the antibody levels of chickens vaccinated with FAV-1 vaccine reached the desired effect.The high antibody titer can be detected from the chickens vaccinated within 4 months,and keeping the medium level in six mouths.These resulte indicated that the vaccine was safe,had long preseved period and good protective efficacy to chickens.
