摘要
目的观察兔角膜爆炸穿孔伤合并海水浸泡后肿瘤坏死因子-α(TNF-α)在角膜组织的表达及其意义。方法成年健康灰兔40只,单支鞭炮致兔角膜爆炸伤。于角膜中周部作3mm的全层切开造成角膜开放伤口。右眼为实验眼,左眼为对照眼。通过角膜切口将海水注入实验眼前房,海水持续灌注眼表30min。对照眼使用生理盐水灌注相同时间。造模后1d、2d、3d、5d、7d采用免疫组织化学法观察角膜组织TNF-α的表达及分布。结果实验组造模后1d角膜上皮细胞及基质中TNF—α表达量最高,以后逐渐降低。实验组角膜组织TNF—α表达造模后1d、2d、3d和对照组相比差异有统计学意义(P〈0.01),造模后5d、7d角膜中TNF—α表达两组比较差异无统计学意义(P〉0.05)。结论兔角膜爆炸穿孔伤合并海水浸泡后早期角膜组织TNF-α的表达明显高于对照组,提示TNF—α参与这一病理改变并发挥重要作用。
Objective To observe the expression of TNF-α in rabbits cornea with explosive penetrating injure and seawater immersion. Methods 40 adult healthy grey rabbits were used for the experiment. The animal model with cornea explosive penetrating injury was developed by using a single firecracker and 3 mm incision in cornea. For each rabbit, the right eye was the experimental eye ( seawater was injected into the anterior chamber through the cornea incision, simulating seawater immersion for 30 minutes) and the left eye was the control eye (the same steps with 0. 9% sodium chloride). Expression of TNF-α in rabbits cornea were detected by immunohistochemistry method on the 1 d, 2 d, 3 d, 5 d, 7 d after the success of modeling. Results Following development of the models, The expression of TNF-α in corneal epithelium and stroma of the experimental group was highest on the 1 st day. There was significant difference in expression of TNF-α between the experimental and control eyes on the 1^st, 2^nd, 3^rd day (P 〈0. 01 ) and difference on the 5^th, 7^th day. Conclusion The expression of TNF-ct in the experimental eye with penetrating cornea injure combined with seawater immersion was significantly higher than that in the control eye, indicating that TNF-α was clearly involved in the pathological change and played an important role in the process.
