摘要
目的提取、纯化鹅不食草植物蛋白,并测定其含量。方法采用硫酸铵沉降法提取鹅不食草植物粗蛋白,通过透析、离子交换层析、分子筛层析等方法分离纯化所得粗蛋白。同时利用SDS-聚丙烯酰胺凝胶电泳法分析蛋白纯化情况,并应用Lowry法测定其含量。结果纯化后的鹅不食草蛋白电泳图谱条带多、清晰,Lowry法测定其蛋白含量高达85%以上。结论鹅不食草植物蛋白的纯化与含量测定方法简便、快捷、准确。为开发利用鹅不食草这一丰富的药用植物资源提供科学依据。
Objective Extraction and purification of Centipeda minima(L.)A.Br.et Aschers.plant protein and detemine its content.Methods Extracted by ammonium sulfate precipitation of crude protein Centipeda minima(L.)A.Br.et Aschers.plants by dialysis,ion exchange chromatography,molecular sieve chromatography,were purified from crude protein.At the same time using SDS-polyacrylamide gel electrophoresis,protein purification tracking the situation,Lowry method Determination of purified protein.Results The purified protein electrophoresis pattern Centipeda bands and more clear,Lowry protein assay was 85%.Conclusion Centipeda minima(L.)A.Br.et Aschers.plant protein purification and determination method is simple,fast and accurate.For the development and utilization of the rich Centipeda minima(L.)A.Br.et Aschers.provide the scientific basis of medicinal plant resources.
