摘要 目的探讨表没食子儿茶素没食子酸酯(EGCG)对酒精性肝病(ALD)小鼠铁过载的作用。方法将24只小鼠随机分为正常对照组、模型组、EGCG治疗组。以酒精灌胃12周制作酒精性肝病(ALD)模型。8周后EGCG治疗组另外给予20mg/kg EGCG腹腔注射治疗。给药4周后处死小鼠,观察各组小鼠肝脏病理变化,并测定肝功能、肝铁含量。结果模型组小鼠血清ALT、AST水平与正常对照组相比显著升高,并且肝组织病理表现肝细胞呈中度脂肪变性,肝铁含量显著升高。EGCG治疗组血清ALT、AST水平显著降低,肝组织病理可见轻度肝细胞脂肪变性,肝铁含量亦显著降低。结论 ALD伴随铁过载,EGCG可以抑制ALD小鼠铁过载,从而发挥其对ALD的保护作用。 Objective To study the effects of Epigallocatechin-3-Gallate(EGCG)on iron overload of mice with alcoholic liver disease(ALD).Methods Twenty-four C57/BL6 mice were randomly divided into three groups: normal group,model group and EGCG treatment group.Mice model of ALD was induced by gavage of alcohol for 12 weeks.At the end of 8 week,EGCG treatment group mice were administered with 20 mg/kg by intraperitoneal injection for 4 weeks.Liver injuries were assessed by histopathologic examination and serum ALT,AST levels.In addition,serum iron levels and liver iron concentration were evaluated.Results Model group mice had marked increase in ALT and AST levels compared with normal group,and their liver tissues showed moderate hepatocytes fatty degeneration.However,EGCG treatments resulted in decreased ALT and AST levels and improved pathological changes.Serum iron levels and liver iron concentration were elevated significantly in model group compared with normal group,but markedly suppressed by EGCG treatments.Conclusion Long-time alcohol consumption may cause iron overload in the liver,and excessive iron exacerbates alcoholic liver injury.EGCG ameliorated iron overload of mice with ALD,the protective effect of EGCG on ALD is associated with its iron-chelating property.
