小RNA干扰降低COX-2表达对乳腺癌细胞增殖和凋亡的影响 被引量:1

摘要 目的:探讨小RNA干扰对MDA-MB-231乳腺癌细胞株COX-2蛋白表达及增殖凋亡的影响。方法:应用已合成的小RNA干扰质粒转染MDA-MB-231细胞株,采用免疫荧光技术和蛋白印迹法(Westernblotting)检测COX-2蛋白的表达;通过MTT实验和流式细胞术分别检测siRNA后对细胞增殖和凋亡的影响结果:转染后72 h,与阴性对照组细胞相比,实验组细胞的COX-2蛋白表达水平明显下降(P<0.01);COX-2减低的乳腺癌细胞的增殖能力比对照组细胞明显受抑制(P<0.05);流式细胞仪检测实验组细胞与阴性对照组细胞相比早期凋亡增加(P<0.05)。结论:利用siRNA技术能够显著抑制乳腺癌细胞株MDA-MB-231中COX-2蛋白的表达;同时能够明显抑制乳腺癌细胞株MDA-MB-231的增殖,并可诱导其凋亡的发生。
