摘要
随着科技的发展.药物相互作用的研究水平有了很大的提高。响应曲面分析法是一种多维模型法.用于描述含有两个以上的自变量(如药物浓度)时.作为因变量(起效时间)的曲面发生变化的规律.是一种既可以定性又可以定量分析药物相互作用的方法。此方法可揭示药物间相互作用的规律.发现曲面中的响应最优区域.寻求药物的最佳配伍方式。以达到指导临床合理用药.降低用药风险和用药成本的目的。本文介绍了响应曲面法的相关概念、模型结构的演变发展及其在临床研究实践中的进展。
With the development of technology, drug interaction study has been greatly improved. Response surface methodology (RSM) is an approach people use to analyze drug interaction both qualitatively and quantitatively; it is a multivariate model of the surface changes of the response/dependent variable (e.g., onset time) over the changes of two or more variables (e.g., drug concentration). This model can be applied to find the desired response region by optimal drugs combination, to explore the mechanism of drug interaction, and thus to provide guidance for clinical rational drug use and reduce the risk and cost. In this article, basic mathematical and pharmacological concepts of the models are introduced, and also the research progress of the clinical application of the method has been reviewed.
