摘要
目的研究麒麟菜海藻色素糖蛋白(SPG)制备及其对小鼠H22肝癌生长的作用。方法在应用溶剂浸提法从麒麟菜中提取SPG,并对样品进行红外线和紫外线光谱扫描、糖肽键分析及多糖和蛋白质的含量测定的基础上,将接种H22肝癌细胞的小鼠分别每天经口灌胃给予高、中、低剂量(100、50、10 mg/kg)的SPG,连续10 d。模型组给予等量的生理盐水。环磷酰胺组隔日腹腔注射环磷酰胺20 mg/kg。检测各组小鼠的瘤质量、体质量、存活率。结果与模型组比较,高剂量SPG组小鼠瘤质量降低,其体质量提高,且差异具有显著性(F=22.54、149.47,q=9.11、29.82,P<0.05),H22肝癌小鼠的存活率明显提高(χ2=5.279,P<0.05)。结论 SPG对小鼠H22肝癌生长具有一定的抑制作用。
Objective To study the inhibitory action of seaweed pigment glycoprotein(SPG) on the growth of liver cancer H22 in mice. Methods SPG was extracted from Eucheuma and scanned by infrared and ultroviolet spectrum and analyzed by cardohydrata-peptide linkage.The mice inoculated with H22 liver cancer cells were divided into high-,moderate-and low-dose SPG groups,which were intragastrically given 10,50 and 100 mg/kg of SGP once a day for 10 days,respectively.To those in model group,an equivalence of normal saline was administered;to those in cyclophosphamide group,an intraperitoneal injection of20 mg/kg of the drug was conducted every other day.The body weigh,survival and tumor weight of the mice in respective groups were recorded. Results Compared with the model group,high-dose SPG reduced tumor weight,increased body weight,the difference being significant(F=22.54,149.47;q=9.11,29.82;P0.05),and raised the survival rate of the mice with liver cancer H22(χ2=5.28,P0.05). Conclusion SPG plays a certain inhibitory role of the growth of liver cancer H22 in the mouse.
