ApoA-I半胱氨酸突变体rHDL对大鼠急性痛风性关节炎影响

EFFECT OF LIPID-BOUND APOA-I CYSTEINE MUTANTS ON ACUTE GOUTY ARTHRITIS IN RATS

摘要 目的探讨载脂蛋白A-I(ApoA-I)半胱氨酸突变体重组高密度脂蛋白(r HDL)对大鼠急性痛风性关节炎的影响。方法健康雄性Wistar大鼠25只,随机分为空白对照组、模型组、卵磷脂组、r HDL74干预组、r HDLwt干预组,每组5只。除空白对照组外其余4组采用右侧踝关节注入尿酸钠晶体诱导急性痛风性关节炎模型,造模后4组分别通过尾静脉注射生理盐水、大豆卵磷脂溶液、r HDL74、r HDLwt进行干预。软尺测量造模后0、2、4、6、8、10 h大鼠踝关节周径;造模10 h后,乙醚麻醉大鼠,内眦取血并取关节腔滑膜组织,ELESA法检测血清中白细胞介素1β(IL-1β)水平,常规检查大鼠关节滑膜的病理学变化。结果与模型组比较,r HDL74能明显抑制关节腔肿胀程度(F=9.61~45.84,q=2.13~16.35,P<0.05),r HDL74及r HDLwt能显著降低血清中IL-1β的水平(F=43.67,q=2.33~16.85,P<0.05);与模型组比较,r HDL74及r HDLwt能明显降低尿酸钠晶体诱导的炎症细胞浸润。结论 r HDL74及r HDLwt对尿酸钠晶体诱导大鼠急性痛风性关节炎有显著的改善作用。 Objective To investigate the effect of lipid-bound cysteine mutants of apoA-I on acute gouty arthritis in rats.Methods Twenty-five male Wistar rats were equally randomized to five groups: control group,model group,lecithin group,rHDL74 group,and rHDLwt group.A rat model of acute gouty arthritis was created by injection of sodium urate crystal(MSU) into the right ankle of the rats,except those in the control group.Upon completion of the models,they were then separately infused with normal saline,soybean lecithin solution,rHDL74,and rHDLwt through vena caudalis into the four groups for intervention.A tape rule was used to measure the circumference of the ankles at 0 h,2 h,4 h,6 h,8 h,and 10 h after the establishment of the models.After 10 hours,blood samples and synovium of articular cavity of the models were taken under ethyl anesthesia.Serum IL-1β level was detected by ELESA,and synovium examined pathologically. Results Compared with the model group,rHDL74 and rHDLwt dramatically inhibited swelling of the rat ankles(F=9.61-45.84,q=2.13-16.35,P0.05),lowered the levels of serum IL-1β(F=43.67,q=2.33-16.85,P0.05),and depressed inflammatory cell infiltration induced by urate crystals. Conclusion rHDL74 and rHDLwt show a remarkable effectiveness in improving symptoms of acute gouty arthritis induced by sodium urate crystals in rats
