厚树脂切片的防脱片方法比较 被引量:1

摘要 目的对厚树脂切片的3种防脱片方法进行比较,探讨哪种防脱片方法更科学实用。方法取甲基丙烯酸树脂包埋的脑组织,切片机40μm厚连续切片,将切片贴在经L-多聚赖氨酸、明胶硫酸铬钾、APES处理过的载玻片上,经过甲苯胺蓝染色后,对防脱片效果进行比较。结果经明胶硫酸铬钾、L-多聚赖氨酸和APES处理的载玻片脱片率分别为3%、10%和13%,明胶硫酸铬钾组防脱片效果明显优于L-多聚赖氨酸组和APES组(u=2.008、2.010,P<0.01)。结论经明胶硫酸铬钾处理防脱片效果好,且更经济,非常适用于厚树脂切片的实验。 Objective To compare three methods for anti-slice escaping with thick resin section,so as to find out the best one for staining process. Methods 40 μm-thick serial sections of the brain tissue embedded by HEMA were taken with a microtome.The slides pretreated with gelatin,Poly-L-Lysine or APES were applied to compare anti-slice escaping effect after toluidine blue staining. Results The ratio of lost sections pretreated with Gelatin,Poly-L-Lysine or APES was 3%,10% and 13%,respectively.The slices pretreated with gelatin were better than Poly-L-Lysine and APES(u=2.008,2.010;P0.01). Conclusion Slices pretreated with gelatin are good,and more economic,which is very applicable to the experiment using a thick resin section.
