BMSCs并BDNF移植治疗大鼠陈旧性脊髓损伤效果 被引量:1

EFFICACY OF COMBINATION TRANSPLANTATION OF BMSCs AND BDNF ON OLD SPINAL CORD INJURY IN RATS

摘要 目的探讨骨髓间充质干细胞(BMSCs)并脑源性神经营养因子(BDNF)移植治疗大鼠陈旧性脊髓损伤的效果。方法对48只Wistar大鼠采用改良的ALLEN撞击法建立脊髓损伤模型,将其随机分为4组,4周后4组分别注射BMSCs+BDNF、BMSCs、BDNF及DMEM干预。分别于移植后1、2、4周采用BBB评分评估大鼠后肢运动情况,并于移植后4周采用免疫组化法检测BMSCs在脊髓损伤部位的分布,及生长相关蛋白43(GAP-43)的表达。结果移植后4周,BMSCs+BDNF组及BMSCs组的BMSCs在脊髓损伤部位均得以存活。BMSCs+BDNF组与BMSCs组及BDNF组比较,脊髓空洞较少并且GAP-43阳性表达面积较高(F=35.57,q=4.97~25.84,P<0.05)。BMSCs+BDNF组BBB评分与其他组比较,差异有显著性(F=27.51,q=3.89~11.57,P<0.05)。结论 BMSCs+BDNF联合移植对于大鼠陈旧性脊髓损伤有较好的修复作用。 Objective To assess the therapeutic effect of combination of bone marrow-derived mesenchymal stem cells(BMSCs) and brain-derived neurotrophic factor(BDNF) transplantation for old spinal cord injury(OSCI) in rats. Methods A model of spinal cord injury(SCI) was created in 48 Wistar rats by using modified ALLEN impacting,which were evenly randomized to four groups.Four weeks later,they were injected with BMSCs+BDNF,BMSCs,BDNF and DMEM,respectively.After one,two and four weeks of therapy,the motor function of hind limbs of the rats was evaluated using BBB scores.The expression of growth-associated protein-43 and the distribution of BMSCs in the injured areas of spinal cord,four weeks after the injection,were measured using immunohistochemical technique. Results After four weeks of transplantation,BMSCs survived in the injured areas of the rats in BMSCs+BDNF group and BMSCs group.Compared with BMSCs and BDNF groups,cavitation was less formed in spinal cord of BMSCs+BDNF group,and with larger area of positive GAP-43 expression(F=35.57;q=4.97-25.84;P0.05).The BBB score of BMSCs+BDNF group was significantly different from that of the other three groups(F=27.51;q=3.89-11.57;P0.05). Conclusion BMSCs+BDNF therapy is better in restoration of old spinal cord injury in the rat.
