摘要
目的探讨阿尔茨海默病病人血浆肿瘤坏死因子转化酶(TACE)活性的检测方法。方法采集临床诊断阿尔茨海默病病人的血液1 mL,抗凝处理后离心获得血浆。分别取2.5、5.0、7.5和10.0μL血浆加入96孔板中,然后加入定量的反应液和酶底物,用荧光酶标仪测定血浆中TACE在37℃与底物的酶反应速度。结果加入7.5μL血浆的反应体系与其他反应体系相比,TACE的酶反应速度最快。结论检测阿尔茨海默病病人血浆中TACE活性的最佳血浆剂量是7.5μL。
Objective To explore the method of detecting the activity of plasma tumor necrosis factor converting enzyme(TACE) in patients with Alzheimer's disease(AD). Methods After anticoagulant treatment,blood plasma was obtained from 1 mL of whole blood collected from AD patient after centrifugation,2.5,5.0,7.5,and 10.0 μL of plasma were added to 96-well plates,respectively.After adding a quantitative reacting buffer and enzyme substrate,the enzymatic activity of TACE was measured by fluorescent microplate reader at 37 ℃. Results A comparison among the four reaction systems,the rate of reaction of TACE was the fastest in reaction system containing 7.5 μL of plasma. Conclusion The optimized dose of plasma to detect the enzymatic activity of TACE in Alzheimer's disease patients is 7.5 μL.
