摘要
目的探讨甲基强的松龙(MP)对马兜铃酸(AA)导致的肾小管上皮细胞损伤的影响。方法 AA组以20mg/L AA刺激近端肾小管上皮细胞株(HK-2),MP组培养体系中加入20mg/L的MP和AA培养48h,另设对照组。透射电镜观察细胞的超微结构,采用MTT法观察HK-2细胞活性,应用流式细胞术检测细胞凋亡百分率。结果与对照组比较,AA组细胞结构发生改变;MP组大部分细胞结构基本正常,部分细胞结构改变明显。与对照组比较,AA组HK-2细胞的活性显著抑制(F=108.522,q=7.215,P<0.01),AA组和MP组凋亡细胞的比例明显增加(F=143.955,q=39.225、23.453,P<0.01);MP组凋亡细胞的比例较AA组明显减少,差异有显著性(q=17.052,P<0.01)。结论 MP通过抑制细胞凋亡起到保护HK-2细胞的作用。
Objective To investigate the influence of methylprednisolone(MP) on impairment of renal tubular epithelial cells induced by aristolochic acid(AA).Methods This study was carried in three groups: AA-stimulation group,20 mg/L of AA was used to activate the cell line of proximal kidney tubules;MP-intervention group,20 mg/L of AA and MP were added and cultured for 48 hours;and control group.A transmission electron microscopy was applied to observe the ultrastructure of the cells;MTT method to measure the activity of HK-2 cells;and a flow cytometry to detect the percentage of cell apoptosis.ResultsCompared with control group,cell structure in AA group was changed;that in MP group was mostly normal,some were changed;the percentage of apoptosis in AA and MP groups increased(F=143.955;q=39.225,23.453;P0.01).The percentage of apoptosis in MP group decreased versus AA group,the difference being significant between them(q=17.052,P0.01).ConclusionMP protects HK-2 cells by inhibiting the apoptosis and raising the resistance of HK-2 cells to AA-induced cell injury.
