消朦灵方对糖尿病大鼠视网膜新生血管的抑制作用研究

Inhibitory Effec of Xiao Meng Ling Decoction on Retinal Angiogenesis in Diabetic Rats

目的探讨消朦灵对糖尿病大鼠的作用及其机制。方法雄性SD大鼠30只,10只未造模作为正常对照组;将造模后成功的20只大鼠随机分为2组:治疗组给予消朦灵方灌胃,模型对照组给予等量温开水灌胃,观察消朦灵方对大鼠全身状态、体重、血糖、SOD、MDA以及视网膜结构的影响。结果与正常对照组比较,模型对照组精神倦怠、萎靡、毛色枯稿无光泽渐呈黄色,甚至脱毛,裸露皮肤,粪便湿稀,量大;治疗组先出现精神倦怠、萎靡、毛色枯稿无光泽呈黄色,打绺,粪便湿稀,量大,继之排尿量逐渐减少,粪便量也逐渐减少。3组大鼠体重均增加,正常对照组增加最多,治疗组次之,模型对照组增加最少,P<0.05,差异具有统计学意义。正常对照组血糖比较稳定,第6周、第10周治疗组的血糖明显低于模型对照组(P<0.05),差异具有统计学意义。治疗组的SOD含量高于模型对照组,差异具有统计学意义(P<0.05);模型对照组的MDA高于治疗组,差异具有统计学意义(P<0.05)。病理切片显示,空白组为正常结构;模型对照组视网膜可见较多新生血管,细胞变形,排列紊乱;治疗组视网膜上新生血管较对照组明显减少,细胞排列较规则,差异具有统计学意义(P<0.05)。结论消朦灵方能减轻糖尿病大鼠部分相关症状,对血糖有一定程度的控制作用,还可以抑制糖尿病大鼠体内的自由基,改善体内氧化-抗氧化平衡状态,预防糖尿病视网膜病变的发生。

Objective To investigate the therapeutic effects and mechanism of Xiao Meng Ling Decoction on diabetic rats.Methods Thirty SD male rats were used for the experiment.Of them,10 rats were in the normal control group,and the other 20 rats were used for the modeling.After modeling,the 20 rats were divided into 2 groups：treatment group were given Xiao Meng Ling by intragastric administration,and the model group received normal saline orally.During the experiment,we observed the effects of Xiao Meng Ling Decoction on rats general state,body weight,blood glucose,superoxide dismutase（SOD）,malondialdehyde（MDA）,and retinal structure.Results Compared with the normal control group,rats in the model group showed spiritual fatigue,low spirits,dull and dry yellow fur,or even hair removal,bare skin,and amount of wet and thin stool.Treatment group first manifested as mental fatigue,low spirits,dull and yellow fur,knotted hair,and amount of wet and thin stool,and then the urine and stool volume decreased.The body weight of the three groups increased,and the increase was obvious in the normal control group,treatment group came next,and the model group was the last（P 〈0.05 compared with that before treatment）.The blood sugar of the normal group remained unchanged,but in the model group the blood sugar was increased,and then was decreased in the treatment group at the 6th and 10 th of the treatment（P 〈0.05 compared with that in the model group）.The treatment group had higher SOD content and lower MDA content than the model group,and the difference was statistically significant（P 〈0.05）.The results of pathological sliced showed that the normal group had normal structure,the model group showed more new vessels with disorded arrangement of deformed cells,and the treatment group had less retinal neovascularization than the control group,and cell arrangement was in order（P 〈0.05 compared with those in the model group）.Conclusion Xiao Meng Ling Decoction can not only relieve partial symptoms and decrease blood glucose,but also inhibit the toxin of free radicals in diabetic rats and improve the balance of oxidation and antioxidation in vivo,thus to prevent the occurrence of diabetic retinopathy.
