益气熄风化痰法对慢性癫痫大鼠大脑海马区GAD65表达的影响

Influence of Methods of Tonifying Qi,Calming Wind and Eliminating Phlegm on Expression of GAD65 in the Hippocampus of Chronic Epileptic Rats Induced by Lithium-pilocarpine

目的观察益气熄风化痰法(痫宁片)对氯化锂联合匹罗卡品所致的慢性癫痫大鼠大脑海马区谷氨酸脱羧酶(GAD65)的免疫反应表达的影响,探讨其抗癫痫的可能机制。方法将70只SD大鼠随机分为空白组、模型组、阳性药对照组和痫宁片组,采用氯化锂和匹罗卡品复制慢性癫痫大鼠模型,分别给予生理盐水、丙戊酸钠、痫宁片灌胃,4周后采用免疫组化SABC法检测大鼠大脑GAD65的表达。结果阳性药对照组与痫宁片组大鼠大脑GAD65表达较模型组增强(P<0.05),痫宁片组与阳性药对照组比较,差异无统计学意义(P>0.05)。结论益气熄风化痰法能增强癫痫大鼠大脑海马区GAD65的表达,这是其抗癫痫的主要机制之一。

Objective To observe the influence of the methods of tonifying qi,calming wind and eliminating phlegm（namely Xianning Tablet） on the expression of GAD65 in the hippocampus of epileptic rats induced by lithium-pilocarpine,and to explore its possible antiepileptic mechanism.Methods Seventy SD rats were randomly divided into blank group,model group,positive control group and Xianning Tablet group.The chronic epileptic rat model was induced by lithium-pilocarpine.The rats were given respectively physiological saline,valproate and Xianning Tablet according to the experimental design.Four weeks later,the expression of GAD65 in the hippocampus of rats was observed by SABC immunohistochemical techniques.Results The expression of GAD65 in the positive control group and Xianning Tablet group was obviously enhanced as compared with model group（P 〈0.05）.There was no statistical significant difference between treatment group and positive control group（P 〈0.05）.Conclusion The methods of tonifying qi,calming wind and eliminating phlegm can obviously enhance the expression of GAD65 in the hippocampus of rats,which may be one of the main antiepileptic mechanisms.
