参青粉对溃疡性结肠炎大鼠HIF-1、VEGF表达的影响 被引量:5

Eeffects of "Shenqing Powder" on hypoxia inducible factor-1 and vascular endothelial growth factor in ulcerative colitis rats

摘要 目的观察参青粉(参三七、青黛)对三硝基苯磺酸(TNBs)诱导的溃疡性结肠炎大鼠结肠黏膜低氧诱导因子-1(HIF-1)、血管内皮生长因子(VEGF)的影响,探讨其作用机制。方法将60只SD大鼠,分为正常组、模型组、SASP组和参青高、中、低剂量组,每组10只。除正常组外,其余各组采用三硝基苯磺酸(TNBs)诱导溃疡性结肠炎模型。造模后第3天开始各组给予相应的干预措施,连续干预7天后处死大鼠。采用免疫组化及Real time-PCR的方法测定各组大鼠结肠黏膜HIF-1及VEGF含量。结果与正常组比较,模型组大鼠结肠黏膜HIF-1和VEGF免疫组化指数、相对mRNA表达升高,差异有统计学意义(P<0.01)。与模型组比较,参青高、中、低剂量组大鼠结肠黏膜HIF-1和VEGF免疫组化指数、相对mRNA表达均下降,差异有统计学意义(P<0.01)。参青各剂量组间比较,高剂量组HIF-1免疫组化指数较中低剂量组升高,差异有统计学意义(P<0.05),而高、中剂量组VEGF免疫组化指数与低剂量组差异有统计学意义(P<0.05);参青高、中剂量组HIF-1和VEGF相对mRNA表达,与低剂量组比较,差异有统计学意义(P<0.05)。结论参青粉能有效下调溃疡性结肠炎HIF-1及VEGF的表达,改善肠道缺氧环境,修复肠道屏障,达到缓解大鼠溃疡性结肠炎的效应。 Objective To observe the effect of "Shenqing p?wder" on hypoxia inducible factor-1 ( HIF-1 ) and vascular endothelial growth factor(VEGF) in TNBsinduced ulcerative colitis rats. Methods Sixty SD rats were randomly divided into 6 groups : normal group, model group, SASP group and Shenqing group ( large, medium, small dose), ten rats in each group. The rat model of ulcerative colitis was induced by TNBs. Each group was treated with 0.9% NACL, SASP and "Shenqing powder" respectively three days after modeling. The rats were sacrificed 1 week later. HIF-1 and VEGF mRNA were detected by Real time-PCFt and immunohistochemisty. Results Compared with the normal group, the levels of HIF-1, VEGF and expression of mRNA in model group were increased significantly ( P 〈 0.01 ), Compared with the model group, the levels of HIF-I, VEGF and expression of mRNA in Shenqing groups ( large, medium, small dose group) were decreased significantly (P 〈 0.01 ). Compared with the medium and small dosage groups, the IHC of HIF-1 in large dose group was increased significantly (P 〈 0.05 ). Compared with small dosage group, the IHC of VEGF in large dosage and medium dosage groups were higher, the difference is significant( P 〈 0.05 ) ; The HIF-I and VEGF had similar effects ;in mRNA, there was a difference between large dose and medium dose groups ( P 〈 0.05 ). Conclusion '" Shenqing powder" has positive effect on ulcerative colitis of rats by decreasing the expression of HIF-1 and VEGF, improving oxygen deficit condition in intestinal tract and repafing intestinal barriers.
