摘要
采用薄层色谱斑点面积法测定了天山大黄中大黄素的含量.样品以乙醇为溶剂,利用索氏提取器提取;在硅胶G薄层板上点样,以甲苯-乙酸乙酯-甲酸(15∶2∶0.98)为展开剂展开.在紫外365nm下检测斑点位置,Rf=0.54,测薄层色谱斑点面积.根据点样量与斑点面积进行线性回归,其回归方程为Y=35.179X+17.65,相关系数R=0.9998;测定出样品中大黄素的含量为1.65%、RSD=0.68%、(n=4).此法所用仪器操作简便、快速、重现性好,为基层单位对该药材的质量评估和药物质量检控提供了快捷方法和依据.
In this paper,the content of emodin in Rheum wittrockii Lundstr.was determined with TLC spot planimetry.The sample was extracted in ethanol with Soxhlet's apparatus;Used silgel G-thin layer plate as the stationary phase,toluene-ethyl acetate-formic acid(15∶2∶0.98) as the mobil phase.The spot was detected at 365nm ultraviolet,with the Rf of 0.54,then the scope was detected.Linear regression was done between the quatity of the sample and the spot scope,with the equation of Y=35.179X+17.65,relevant factor R=0.9998.As a result,the content of emodin in Rheum wittrockii Lundstr is 1.65% and RSD 0.68%、(n=4).This method has the advantage of convenient,fast,good recurrence,which provides a simple method and a reliable proof to quality evaluation and examination for the primary level medication department.
