半滑舌鳎幼鱼消化酶同功酶谱初步研究 被引量:1

摘要 以半滑舌鳎幼鱼为研究材料,利用聚丙烯酰胺非变性电泳和同功酶活性染色对半滑舌鳎消化酶进行了初步分离。蛋白酶采用明胶原位消化法,肝脏蛋白酶检测出3条酶带,其中1条强带,2条弱带;后肠蛋白酶出现了2条酶带,其中有1条强带,1条弱带,并且与肝脏蛋白酶的前2条酶带分别在同一分子量水平上;前肠和中肠蛋白酶均只出现了1条弱带,且在同一分子量水平上。半滑舌鳎肠道各部分的淀粉酶带都在相似分子量附近出现了一条弱酶带,而肝脏在不同的分子量水平出现了1条酶带。肝脏、前肠和后肠脂肪酶均出现了4条酶带,其中,肝脏和前肠均分别有1条强带,1条中强带,2条弱带;后肠脂肪酶则有1条中强带,3条弱带;中肠脂肪酶出现了3条弱带。肝脏和肠道各部分脂肪酶的第一条酶带都在同一分子量水平上,而且活性相似。 The digestive enzymes in juvenile tongue-sole Cynoglossus semilaevis were separated and analyzed through acrylamide gel electrophoresis.(1)Glutin digestion in situ was applied to protease.Liver protease was composed of three bands,one bright band and two dim bands.Midgut protease and foregut protease included one dim bands,which paralleled liver protease's dim band.Hindgut protease was composed of one bright band and one dim band and the dim they had similar molecular weight with the liver protease.(2)Isozyme technology was applied to amylase.Foregut,midgut and hindgut amylase all had one dim band.Liver amylase had one dim band which had different molecular weight with the intestines. (3)Isozyme technology was still used to lipase.Four bands were found from foregut,hindgut and liver lipase while three bands were found from midgut.Their first bands all had similar molecular weight.
