摘要
目的 探讨转化生长因子 - β1(TGF - β1)和血管内皮生长因子 (VEGF)在大肠癌中的表达以及TGF - β1和VEGF表达与肿瘤内微血管密度 (MVD)的关系。方法 应用免疫组织化学SP法检测 6 0例大肠癌中TGF - β1、VEGF的表达及MVD值 ,并以 10例大肠腺瘤和 10例正常大肠黏膜组织作对照 ,分析大肠恶性肿瘤组织中TGF- β1与VEGF的表达与MVD的关系。结果 6 0例大肠癌中 ,TGF - β1阳性 14例 (2 3.3%) ,VEGF阳性 2 1例(35 %) ,MVD为 5 0 .7± 9.49。TGF - β1表达与肿瘤的分化程度、浸润深度、临床分期和生存率有关 ,与VEGF表达、MVD呈明显正相关 (P <0 .0 5 )。结论 TGF - β1是大肠癌血管生成的重要因子 ,其表达与细胞本身分化程度有关 ,可通过高调控VEGF的表达促进血管生成 ,是有价值的预后因素。
Objective To study the expression of transforming growth factor beta 1 (TGF-β1) and vascular endothelial growth factor (VEGF) in the tissue of colorectal cancer and the relationship with microvessel density (MVD). Methods The expression of TGF-β1 and VEGF were detected with immunohistochemical SP method in 60 cases of colorectal cancer and the value of MVD were measured simultaneously. 10 cases of colorectal adenoma and 10 cases of normal colorectal mucosae were used as control. The relationship between expression of TGF-β1 and VEGF with MVD was evaluated. Results TGF-β1 and VEGF expression were detected in 14 (23.3%) and 21(35%) of 60 respectively, and the MVD was 50.7±9.49. TGF-β1 expression was significantly correlated with the histological type, the depth of invasion, the stage of disease and five year survival rate. There was a close correlation between TGF-β1 expression and VEGF expression. There was no correlation between TGF-β1 expression and MVD, whereas VEGF expression was significantly correlated with MVD. Conclusions The expression of TGF-β1 has close relation with the differentiation of cancer cell, and might be associated with tumor progression by indirectly stimulating angiogenesis through the up-regulation of VEGF expression in colorectal cancer.
