摘要
目的:探讨人骨髓在液氮长期保存下造血细胞活力.方法:以10%二甲基亚砜、“二步”冻存法在液氮液相中冻存人骨髓细胞,用造血细胞培养的集落形成为指标,定期观察造血细胞活力.结果:20份人骨髓,1~15年CFU-GM回收率由(72.4±10.3)%下降至(40.5±13.2)%,CFU-GM回收率相关公式为:Y=0.7822-0.02573X.15年后BFU-E、CFU-GEMM、LTC-IC形成能力良好.结论:在液氮液相中冻存入骨髓,15年后仍有较好的造血细胞活力.
Objective: To observe the effect of cryopreservation on activities of human bone marrow cells. Method: 10% dimethylsulfonamide was used as cryoprotectant and cooling procedure was performed with two-step method. Bone marrow samples were stored in liquid phase of liquid nitrogen. Hematopoietic activities were assayed by colony-forming units of hematopoietic progenitors. Results: After 15 years preservation, the CFU-GM recovery rate was decreased from (72. 4±10. 3%)% in 1st year to (40. 5±13. 2)% in 15 th year. The counted regression equation of the CFU-GM recovery rate is Y = 0. 7822 - 0. 02573X. There remains to be enough production of BFU-E, CFU-GEMM, LTC-IC after 15 years. Conclusions: The capacities of bone marrow cells to produce colony-forming units can be protected by liquid nitrogen cryopreservation as long as 15 year.
