中国汉族慢性乙型肝炎患者MBP基因多态性特点及意义

摘要 目的 比较了汉族人群中慢性乙型肝炎患者与背景对照组、健康对照组之间甘露糖结合蛋白(MBP)基因多态性的差异。方法 应用PCR-RFLP方法对中国汉族的552例样品进行MBP基因多态性进行检测,并将其中的22例样品的PCR产物直接进行DNA序列分析。结果 中国汉族人群中慢性乙型肝炎患者与背景对照组、健康对照组之间MBP基因突变频率分别为0.174、0.182和0.202,未检测到密码子52和57的基因突变。结论 MBP基因密码子54等位基因多态性在中国汉族慢性乙型肝炎患者、背景对照组与健康对照组中的分布均符合Hardy-Weinberg平衡,而且各组之间MBP基因突变频率没有显著差别。 Objective To detect the genotypes of the exton 1 of human mannose-binding protein (MBP) allele of Chinese Han chronic hepatitis B patients. Methods Genotypies of MBP gene from 552 samples of Chinese Han Hepatitis B patients, background controls and health controls were detected by PCR-RFLP. The exon 1 of MBP gene was analysed using ABI 310 genetic analyzer of 22 samples. Results Allele frequencies of the codon 54 variant of Chinese Han hepatitis B patients, background controls and health controls respectively were 0. 174,0. 182 and 0. 202. The allele mutations of the codon 52 and 57 were not detected. Conclusion Allele distribution for the codon 54 mutation of Chinese Han hepatitis B patients, background controls and health controls was in good agreement with Hardy-Weiberg equilibrium. There were not differences of the allele frequencies between chronic hepatitis B patients, background controls and health controls.
