摘要
目的 寻找一种简便准确的方法来评估供肝质量。方法 建立原位隔离冷灌注模型模拟整个肝移值过程,观察复流后细胞周期变化情况,用预处理的手段获得不同损伤程度的供肝模型,复流后比较反映周期变化的指标(增殖细胞核抗原)和反映肝功能的指标(AST、ALT、LDH)。结果 复流后肝脏增殖细胞核抗原的表达量显著增高,G_1期缩短至2小时左右,整个细胞周期缩短至20小时左右;增殖细胞核抗原的表达量与供肝损伤程度在一定范围内存在显著的相关性。结论 增殖细胞核抗原的表达可作为判断供肝质量状况的一个有效指标。
Objective To look for a simple exact method to evaluate the quality of liver as graft.Methods The simple total process of liver transplantation is acquired by developing model of isolated liver cold perfusion(ILCP). Then observe the change of liver cell cycle. Corresponding injury level liver is made through different kind of preconditioning. Compares index of changes of cell cycle (PCNA)and that of liver function (AST、ALT、LDH). Results Parts of liver cells move forward proliferating statue promptly. The phase G1 is shorted to 2hours while the total cycle is to 20 hours. The index of PCNA is potently related to liver function in some extent. Conclusion PCNA may be seen as an effective index to evaluate liver function.
