VEGF-C/VEGFR-3与肿瘤淋巴结转移研究进展 被引量:7

摘要 恶性肿瘤治疗失败的原因是肿瘤细胞扩散和转移.血管内皮生长因子(vascular endothelial growth factor,VEGF) 家族是唯一特异性作用于脉管内皮细胞的生长因子,是一组分泌性糖蛋白,包括VEGF-A( 即以往发现的VEGF) 、VEGF-B 、VEGF-C 、VEGF-D 、VEGF-E 、P1GF( 胎盘生长因子)和PDGF( 血小板衍生生长因子)7 大成员.它们在肿瘤组织血管内皮细胞、淋巴管内皮细胞的分裂增殖过程中起关键作用.血管内皮生长因子-C(vascular endothelial growth factor-C,VEGF-C) 首次由Joukov 在1996 年报道以来,其作为一种特异性的的血管、淋巴管内皮细胞调节因子,通过与其受体血管内皮生长因子受体-3(vascular endothelial growth factor receptor,VEGFR-3) 结合后,能诱导血管及淋巴管内皮细胞生长,在肿瘤组织中表达增加,促进实体瘤内淋巴管的新生, 促进肿瘤细胞进入淋巴管并通过淋巴系统向全身扩散.应用相应的抗体阻断VEGF-C 与VEGFR-3 的结合,可起到抑制淋巴管新生的作用.现将VEGF-C/VEGFR-3 与肿瘤淋巴结转移研究进展综述如下.
