摘要
目的研究不同亚型基因工程α-干扰素对大鼠肝贮脂细胞增殖和胶原合成的影响。方法分离、培养大鼠肝贮脂细胞,采用 MTT 法和~3H-脯氨酸掺入法分别测定细胞增殖情况和胶原合成情况。结果α1b、α2b 基因工程干扰素在1000~10000 U/ml 度范围内,能显著抑制贮脂细胞的增殖及胶原合成,其作用呈剂量浓度依赖性。结论国产的基因工程α1b、α2b 干扰素能显著抑制大鼠肝贮脂细胞增殖和胶原合成,提示对肝纤维化有直接防治作用。
Objective To investigate the effect of interferon-α on proliferation and collagen synthesis of rat hepatic stellate cells. Methods MTT and ~3H-proline incorporation methods were conducted respectively to assay the parameters.Results The inhibition effect on both cell proliferation and collagen synthesis was observed by either interferon α-1b or interferon α-2b,and there was no obvious difference in action befween the two subtype interferon action.Conclusion The domestically produced interferon-α has an antihepatofibrogenesis potential.
