摘要
用6 0 Co -γ射线照射侧耳P3 2 孢子 ,得到精氨酸营养缺陷型的突变体[1]。突变体的原生质体与经 0 83 %碘乙酰胺灭活处理的美味侧耳单核菌丝释放的原生质体进行融合 ,然后涂布在基本再生培养基上 (RMM) ,得到 5个融合子菌落 ,选其中 1个菌丝生长浓密的菌落进一步做出菇试验。经酯酶同工酶分析表明 ,融合菌株具有 7条酶带 ,其中有 3条酶带与侧耳P3 2 相同 ,2条酶带与美味侧耳相同 ,还出现了 2条双亲没有的酶带 ,证实为双亲融合杂种。融合菌株的子实体性状继承了双亲的遗传基础 ,氨基酸含量高于双亲 。
The Arg - mutations were obtained by radiating the spores of pleurotus p 32 with C o60.The protoplasts of mutants and the protoplasts of uninucleate mycelium of pleurotus sapidus treated by 0.83% CH 2ICONH 2 were mixed.Then spread the mixture on the RMM.Five hybirds were obtained.Among the five,choose one growing best to make tests. Analysis of it with esterase isozymes zymograms proved that among the seven bands of fus-ants,three shared the same bands with pleurotus P 32 ,two were the same as that of Pleurotus sapidus,and two were new bands.It was proved that the fusant was a hybrid of two parents.The characters of mushroom inherit the genetic basis of the two parents.But its content of amino acid is higer than its parents'. Hence the hybird has certain heterosis.
