转γ-干扰素基因大鼠肝细胞模型的建立

The Construction of Murine Hepatic Model Transferred with Interferon γ Gene

目的：使用多聚阳离子脂质体载体（ｌｉｐｏｆｅｃｔＡＭＩＮＥ）在体外建立转γ干扰素（ＩＦＮγ） 基因的大鼠肝细胞模型。方法：利用ｌｉｐｏｆｅｃｔＡＭＩＮＥ在体外将ＩＦＮγ基因转染大鼠肝细胞。分别以ＲＴＰＣＲ 和ＥＬＩＳＡ方法检测所转ＩＦＮγ基因在大鼠肝细胞的转录和表达情况。结果：以ｌｉｐｏｆｅｃｔＡＭＩＮＥ为载体的ＩＦＮγ基因转染大鼠肝细胞后，可被有效的转录并产生具有生物活性的ＩＦＮγ。结论：ＩＦＮγ基因可被成功转染入大鼠肝细胞并可有效表达。这可能为细胞因子的免疫治疗提供一种新途径。

Objective:To investigate interferon γ (IFN γ) gene transfer and expression in BRL murine hepatocyte with lipofectAMINE vector in vitro. Methods:IFN γ gene was transferred and expressed in BRL murine hepatocyte with lipofectAMINE vector in vitro. Its mRNA transcription and protein concentration were determined by RT PCR and ELISA assay respectively. Results:The IFN γ gene that was transferred with lipofectAMINE vector in vitro can be transcripted in the BRL murine hepatocyte continuously.Conclusion:IFN γ can be transferred successfully and expressed efficiently in BRL murine hepatocyte.This may lead to a new therapeutic methods for cytokine immunological therapy.
