摘要
目的:使用多聚阳离子脂质体载体(lipofectAMINE)在体外建立转γ干扰素(IFNγ) 基因的大鼠肝细胞模型。方法:利用lipofectAMINE在体外将IFNγ基因转染大鼠肝细胞。分别以RTPCR 和ELISA方法检测所转IFNγ基因在大鼠肝细胞的转录和表达情况。结果:以lipofectAMINE为载体的IFNγ基因转染大鼠肝细胞后,可被有效的转录并产生具有生物活性的IFNγ。结论:IFNγ基因可被成功转染入大鼠肝细胞并可有效表达。这可能为细胞因子的免疫治疗提供一种新途径。
Objective:To investigate interferon γ (IFN γ) gene transfer and expression in BRL murine hepatocyte with lipofectAMINE vector in vitro. Methods:IFN γ gene was transferred and expressed in BRL murine hepatocyte with lipofectAMINE vector in vitro. Its mRNA transcription and protein concentration were determined by RT PCR and ELISA assay respectively. Results:The IFN γ gene that was transferred with lipofectAMINE vector in vitro can be transcripted in the BRL murine hepatocyte continuously.Conclusion:IFN γ can be transferred successfully and expressed efficiently in BRL murine hepatocyte.This may lead to a new therapeutic methods for cytokine immunological therapy.
