摘要
目的探讨在腹主动脉瘤形成过程中内源性蛋白酶与动脉中膜层弹力纤维降解的关系。方法18条犬随机分为两组,生理盐水对照组和胰蛋白酶造模组各9条。分别于第7、30、90d处死实验犬并获取主动脉标本,用组织化学方法观察动脉壁基质成分改变,用底物凝胶电泳分析内源性蛋白酶表达。站系实验组主动脉直径于30d后可见扩张,弹力纤维明显减少,观察到多种内源性蛋白酶表达并于90d表达更为明显,同时有炎性细胞浸润。给俺胰蛋白酶注人腹主动脉夹闭段后诱导内源性蛋白酶的出现,通过一系列复杂的细胞和生化机制,降解弹力纤维,导致主动脉扩张形成动脉瘤。
Objective To study the relationship between endogenous proteinase and thedeclining elastin content of the aortic media during the development of abdominal aorticaneurysm(AAA). mthods Nine dogs were perfused with the test solution to the abdominalaorta, and nine with NS. Laparotomy was performed on day 7,30,90 to harvest the dilatedaorta. Histochemical studies were performed to analyze the matrix opponents and substrategel enzymography was used to observe the appearance of endogenous proteinase. Rcs- ults Elastin was apparently degenerated from the first month and the aorta dilatedgradually. From the first month mutiple endogenous matrix metalloproteinase becamedetectable. Histologic study revealed infiltration of the aorta with inflammatory cells. ConcIusio' The results suggest perfusion of trypsin can induce endogenous metal-loproteinase, which degrades elastin to develop aneurysm-
