摘要
目的用激光共聚焦显微术探讨缺氧过程中心肌细胞损伤的离子机制。方法取大鼠心脏分离心肌细胞,实验组和对照组各20份,分别在含磷酸肌酸和无磷酸肌酸的培养基中孵育24~72h,fluo3AM染色,488nm激光激发,选择固定视野以33ms间隔记录3min,分析荧光强度变化规律。结果标本荧光呈周期性变化,对照组周期长,且不稳定,实验组明显缩短,周期稳定。结论磷酸肌酸具有对缺氧心肌的保护作用。
Purpose To ascertain the ionic mechanism of myocardial injury after by means of confocal
laser scanning microscope. Methods Myocardial cells,which were derived from rat
myocardium dissociated with collagenase and hyaluronidase,were cultivated in the medium
either with or without creatine phosphate(CP) for 24 to 72 hours,during which a hypoxic
environment was made in the first 5 hours.After 24 or 72 hours incubation myocardial cells were
loaded with flou 3AM,a Ca 2+ marker,and observed under a confocal laser scanning
microscope . Results The fluorescence intensity was showing a rhythmical change.The
frequency of the rhythm was stable and high in the specimen after 72 hours cultivation and in
the CP group it became stable earlier and higher than that of the control group. Conclusions
Creatine phosphate proves myocardial protective and antiarrythmic.
