摘要
运用40个10碱基随机引物对长江中游鲢的遗传多样性进行了随机扩增多态DNA (RAPD)分析。所用引物中,能在鲢不同个体基因组中扩增出多态DNA带的有10个,占总引物数 的 25%.据RAPD带型分析,所取长江鲢自然群体的 18条样品中,每两个体相比较的遗 传相似度在0.9285至0.9788之间,平均值为0.9543;遗传变异度则在0.0212至0.0715之 间,平均值为0.04725;群体Shannon表型多样性指数(Ho)为8.8615,Shannon多样性值(H)为0.0624。 讨论了这些数据所反映的长江鲢遗传多样性和鱼类种质资源评价问题。
Genetic diversity analysis of silver carps (Hypophthalmichgthys molitrix ) from Yangtze River was dried out by RAPD technigue using 40 random oligonuclcotide primers (10 mer each). Total 142 bands amplified with 36 effective primers were shown on electrophoretogram and only 10 of total 40 primers (25%) produced polymorphic bands in different individuals. Based on RAPD data, the genetic similarity (S) and variability diversity (D) between individuals in the population are 0.9543 (S) and 0.04725(D)on avers, ranging from 0. 9285 to 0. 9788 and from 0. 0212 to 0.O715 separately. The Shannon's index (Ho) and value (H) of phenotypic diversity of the population is 8. 8615 (Ho) and 0. 0624 (H) respectively. The genetic diversity of silver carp in Yangtae River and RAPD mark application in evaluation and management of fish resource were discussed.
