鸡败血霉形体抗原基因的筛选和DNA电泳鉴定

Screening and Determination of Mycoplasma gallisepticum Antigen Gene by DNA Electrophoresis

应用鸡败血霉形体抗血清筛选鸡败血霉形体Ｓ６株基因文库，得到１７９个阳性克隆，不足重组噬菌体的１％。反复筛选后，获得５个强阳性克隆。抽提ＤＮＡ酶切鉴定，发现２个克隆的外源片段在５ｋｂ左右，另２个在２．５ｋｂ左右，一个为３．６ｋｂ，为特异抗原的制备和基因工程疫苗的研究打下良好基础。

gtll positive clones, less than 1% of the recombinant were selected by screening a M. gallisepticum (MG) genomic DNA library with antisera against MG cells and their membrane proteins. After repeated screening, 5 strong positive clones were selected, 2 clones showing to have about 5 kb inserts, 2 clones showing to have about 2.5 kb inserts and 1 clone showing to have a 3.6 kb insert by electrophoresis. This study may serve as the basis for a vaccine against MG infection and preparation of specific antigen.
