RG50864对PDGF诱导的PASMC DNA含量及增殖细胞核抗原表达的影响

摘要 目的:研究酪氨酸蛋白激酶抑制剂RG50864对PDGF诱导的肺动脉平滑肌细胞(PASMC)DNA含量及增殖细胞核抗原表达的影响。方法:应用原代培养的小牛肺动脉平滑肌细胞,用流式细胞仪分析RG50864对PDGF诱导的PASMCDNA百分含量及增殖细胞核抗原的变化。结果:与对照组相比,RG50864处理组可显著地抑制PDGF诱导的PASMC增殖细胞核抗原表达的荧光强度及荧光指数(P<0.001),增加PASMC生长周期中的G0/G1期的DNA百分含量,抑制S期及G2M期DNA百分含量(P<0.01或P<0.001)。结论:RG50864可显著地抑制PDGF诱导的PASMC增殖细胞核抗原表达,抑制PASMC周期S期及G2M期DNA百分含量,阻止PASMC周期由G0/G1向DNA合成的S期及G2M期转化。 The study was designed to explore the effects of RG50864 on DNA percent and proliferation cell nuclear antigen (PCNA) of PDGF induced pulmonary artery smooth muscle cells (PASMC).Methods:By primary culture of calf PASMC,the changes of RG50864 on DNA percent content and PCNA of PDGF induced PASMC were observed by using flow cytometry.Results:As compared with control group,fluorescence intensity and index of PDGF induced PASMC PCNA expression were significantly inhibited by RG50864 groups ( P<0.001 ),DNA percent content of G 0/G 1 phase was increased,however,which of S and G 2M phase was decreased in RG50864 groups ( P <0.1 or P<0 001 ).Conclusion:RG50864 could significantly inhibite the PCNA expression of PDGF induced PASMC,and block the converting of cells from G 0/G 1 phase to S phase of DNA synthesis.
