摘要
目的:了解普萘洛尔对豚鼠皮肤的环核苷酸(CN)及组织学的影响。方法:参照黄畋、Gaylarde等采用普萘洛尔乳剂外涂豚鼠背部皮肤,用放射免疫分析法测定表皮CN,还观察了组织学变化。结果:造成了银屑病样病理变化,且表皮内CN含量变化,模型组表皮内cAMP下降、cGMP升高及其比值cAMP/cGMP下降,与正常组比较有极显著性差异(P<0.01)。结论:证实表皮内CN变化是引起皮肤增殖活化的因素之一。为探讨银屑病模型及用作表皮增殖动力学、药物筛选提供依据。
Objective:To observe the effects of propranolol on levels of cyclic nucleotides and histology of the back skin of Guineapigs.Methods:Propranolol emulsion was topically applied to the back skin that the hair has been removed according to methods described by Huangmei, Gaylarde, et,al. Skin cyclic nucleotide contents were determined by radioimmunoassay(RIA) and skin histology changes were also examined.Results:Propranolol caused a psoriasiform change of epidermis of the guineapigs and a increase in cyclic guanosine monophosphte(cGMP), decrease in cyclic adenosine monophosphate(cAMP) and therefore decrease in cAMP/cGMP ratio, with a highly significant difference between propranololtreated group and the control (P<0.01).Conclusion:It is demonstrated that the changes of cyclic nucleotide contents is one of factors causing proliferation of skin and, this may provide a pathological model for kinetic study of skin proliferation and for screening drug for psoriasis.
