摘要
目的探讨依达拉奉对帕金森病(PD)小鼠黑质多巴胺神经元的保护作用。方法 30只健康雄性C57BL/6小鼠,随机分为正常组、模型组和治疗组。模型组和治疗组给予腹腔注射百草枯和代森锰建立PD模型,建模成功后,治疗组给予依达拉奉腹腔注射2 w。观察3组小鼠行为学的改变、并用免疫组织化学法检测黑质TH、GSK-3β的表达。结果模型组与治疗组比较,爬杆时间明显延长,黑质TH阳性神经元数量显著降低,GSK-3β的表达显著增高。结论依达拉奉治疗可显著改善百草枯和代森锰引起的损伤,有明显的神经保护作用。
Objective To investigate the protective effect of edaravone on nigrostriatal dopaminergic neurons in Parkinson′s disease(PD) mice induced by paraquat and maneb.Methods Male C57BL/6 mices were randomly divided into paraquat(PQ) + maneb(MN) treated,edaravone treated and control groups.Behavioral and immunohistochemical changes were to detect the motor coordination and the expression of tyrosine hydroxylase(TH) and GSK-3β in the substantia nigra pars compacta(SNc).Results Edaravone significantly attenuated the impairment of behavioral performance,increased the number of TH positive neurons and decreased the expression of GSK-3β in SNc.Conclusions Edaravone has a beneficial influence on PD induced by PQ+MB.
