摘要
在低氧条件下,猪肺动脉内皮细胞(PAEC)对猪肺动脉平滑肌细胞(PASMC)增殖调控,是研究低氧性肺动脉高压时肺动脉结构重组的重要内容。应用MTT比色法、DNA荧光测定法,观察了低氧PAEC条件培养基(HECCM)中多胺促PASMC增殖的作用。结果表明,在PAEC培养液中加入鸟氨酸脱梭酶抑制剂二氟甲基鸟氨酸(DFMO),可阻止HECCM促PASMC增殖的效应。故认为低氧刺激PAEC产生的多胺亦是HECCM中促PASMC增殖的一个重要因子。
PASMC proliferation regulated by PAEC is very important in pulmonary vascular remodelingunder the circumstance of hypoxial pulmonary hypertension. Basing on that exogenous Polyamine could promote the proliferation of PASMC and that DFMO could thoroughly inhibit this process,we studied the effectof Polyamine within HECCM on the PASMC proliferation. The results showed that DFMO could inhibitPASMC proliferation which was induced by Polyamine in HECCM. Thus it demonstrated that Polyarnine wasan important factor of PASMC proliferation stimulated by HECCM. This study provides a basis on which thecomplicated interactions between PAEC and PASMC can be further investigated.
