Expression of Aurora-B and Ki-67 in osteosarcoma and its correlation with distant metastasis

摘要 目的探讨Aurora-B、Ki-67在骨肉瘤中的表达及与远处转移的关系。方法收集2002年1月-2009年3月手术切除的60例骨肉瘤组织石蜡标本,采用免疫组织化学方法检测Aurora-B和K-i67的表达,以同期21例骨软骨瘤组织作为对照。结果 Aurora-B蛋白在骨肉瘤组织中的阳性率(53.3%)明显高于骨软骨瘤(14.3%,P<0.05),且在伴远处转移的骨肉瘤组织中(78.6%)明显高于无远处转移者(45.7%,P<0.05)。K-i67增殖指数在骨肉瘤组织中为35.1%±15.2%,明显高于骨软骨瘤组织(2.1%±0.9%,P<0.01),且在伴远处转移的骨肉瘤组织中(48.3%±20.1%)明显高于不伴远处转移者(21.2%±10.2%,P<0.05)。相关分析显示,骨肉瘤组织中Aurora-B和Ki-67蛋白表达呈显著正相关(r=0.885,P<0.01)。结论 Aurora-B和Ki-67蛋白可能与骨肉瘤的发生及远处转移有关,有望成为骨肉瘤新的诊断及分子治疗靶点。 Objective To investigate the correlation of the expressions of Aurora-B and Ki-67 protein in osteosarcoma,and the relationship of the expressions with distant metastasis.Methods Sixty formalin-fixed,paraffin-embedded surgical specimens of osteosarcoma obtained from Jan.2002 to Mar.2009 were used to observe the expressions of Aurora-B and Ki-67 by immunohistochemical staining,and 21 specimens of osteochondroma obtained at the same period were similarly studied to serve as control.Results The positive rate of Aurora-B was significantly higher in osteosarcoma(53.3%) compared with osteochondroma(23.8%,P〈0.05),and was markedly higher in osteosarcoma with distant metastasis(78.6%) than in osteosarcoma without distant metastasis(45.7%,P〈0.05).The Ki-67 proliferation index was significantly higher in osteosarcoma(35.1%±15.2%) than in osteochondroma(2.1%±0.9%,P〈0.01),and was markedly higher in osteosarcoma with distant metastasis(48.3%±20.1%) than in osteosarcoma without distant metastasis(21.2%±10.2%,P〈0.05).Correlation analysis revealed that the Aurora-B positive rate was positively correlated with Ki-67 proliferation index(r=0.885,P〈0.01) in osteosarcoma.Conclusion Aurora-B and Ki-67 protein may be involved in the development,progression and metastasis of osteosarcoma,and is expected to serve as a new diagnostic and therapeutic target for osteosarcoma.
