胎肝无细胞悬液对脐血CD34^(+)细胞体外扩增的研究

A STUDY OF THE IN VITRO EXPANDING EFFECT OF FETAL LIVER CELL-FREE ON UMBILICAL CORD BLOOD CD34^+ CELLS

采用两步法分离出脐血CD34＋细胞，比较研究了胎肝无细胞悬液及IL3＋IL6＋GM－CSF＋EPO两种情况下脐血CD34＋细胞的体外扩增。结果表明胎肝无细胞悬液对粒系-单极系祖细胞（Granulocyte－macrophagecolony－formingunit，CFU－GM），红系暴式集落形成单位（Burst－formingunitoferythroid，BFU－E），多向祖细胞（Mixedcolony-formingunit，CFU－MIX）3种集落的扩增效果明显优于4种造血因子联合组，差异具有统计学意义（P＜0.01）。脐血造血细胞扩增对于成人脐血移植有重要意义。胎肝无细胞悬液的扩增成功表明其可代替多种造血生长因子联合应用，并可作为一种效果优良、价廉的扩增剂。

Using two-step-adhering method we seperated CD34+ cells from human umbilical cord blood and compared the expanding effect of fetal liver cell-free and IL3+IL6 +GM -CSF+EPO on umbilical cord blood CD34+ cells. The results showed that the expanding effect of fetai liver cell-free on CFU-GM, BFU-E, CFU-MIX was superior to that of combination of several hematopoietic growth factors. The difference has statistical value (P＜0.01).In vitro expansion is important to adult in umbilicalcord blood transplantation.The successful expan sion of fetal liver cell-free shows that fetal liver cell free may replace the combination of many kinds of hematopoietic growth factors. As an expander more effective and cheap fetal liver cell-free will be good for mankind.
