摘要
本文进行了不同离子交换剂的选择及静态和动态离子交换纯化蚓激酶的工艺条件研究。结果表明,选用DEAE-纤维素为纯化蚓激酶的离子交换剂,以0.02mol/L,pH7.8磷酸缓冲液+0.5mol/LNaCl作为洗脱剂,蚓激酶纯化效果较好。蚓激酶比活达3.25×104u/mg,比口服用蚓激酶活力提高了3.6倍。
The optimum conditions of Lumbrokinase by ion-exchange chromatography were studied . As a result, while DEAE-cellulose (D52) and 0.02 mol/L, pH7.8 phosphate buffer + 0.5 mol/L NaCl solution were selected to be eluent, the purification efficiency was bette. Lumbrokinase activity was up to 3.25 × 104 u/mg, was as 3 .6 times high as the achivity of Lumbrokinase for orally taken.
