5-FU联合胸腺肽对H_(22)细胞皮下移植瘤小鼠免疫功能的影响 被引量:3

5-FU combined with thymopentin injection inhibit growth of hepatocellular cancer by regulating immune fuction

摘要 目的:探讨5-FU联合胸腺肽的抗肿瘤及免疫调节功效。方法:32只ICR小鼠建立H22小鼠肝癌细胞的皮下移植瘤模型,分为对照组(生理盐水),5-FU组(10mg/kg),5-FU+胸腺肽组(10mg/kg+0.2mg/kg),胸腺肽组(0.2mg/kg),连续给药5天后测量肿瘤重量和体积,检测外周血WBC及淋巴细胞(LY)数量及外周血中CD3+、CD4+、CD8+、NK细胞的百分含量、血清中白介素2(IL-2)和肿瘤坏死因子(TNF-α)的浓度。结果:5-FU联合胸腺肽显著抑制了小鼠皮下移植瘤的生长(P<0.01),胸腺肽上调了5-FU降低的WBC及LY细胞数量(P<0.05),增加外周血中CD3+、CD4+及NK细胞的水平(P<0.05);与对照组比,显著升高了血清中IL-2和TNF-α的浓度(P<0.01)。结论:5-FU联合胸腺肽可增强机体的细胞免疫功能,上调血清中细胞因子的表达水平,具有显著的抗肿瘤功效。而不良反应较小。 Objective:To study the inhibitory effect of 5-FU and thymopentin injection on hepatocellular cancer(HCC),and investigate the mechanism mediating its bioactivity.Methods: H22 tumor-bearing ICR mice were treated with normal saline(NS),5-FU(10mg/kg),thymopentin injection(0.2mg/kg),5-FU+thymopentin injection(10mg/kg+0.2mg/kg)for 5 days.Tumor volume and tumor weight,percentages of CD3+,CD4+,CD8+ and NK cells in peripheral blood,the number of WBC and LY,and serum levels of IL-2 and TNF-α were evaluated.Results: 5-FU and thymopentin injection produced a potent anti-tumor effect(P0.01).Mice treated with 5-FU and thymopentin injection had higher percentage of CD3+,CD4+ and NK cells(P0.05)in the peripheral blood than treated with NS.Mice receiving 5-FU and thymopentin injection had the higher number of WBC and LY(P0.05),and serum levels of IL-2 and TNF-α(P0.01).Conclusion: 5-FU and thymopentin injection can inhibit HCC growth by regulating immune function.
