粉防己碱对宫颈癌细胞周期影响及其机制研究

Effects of tetrandrine on cell cycle regulatory processes of HeLa cells and their mechanisms

目的:研究粉防己碱对HeLa细胞的抗肿瘤作用,并探讨其分子机制。方法:用MTT法检测粉防己碱对HeLa细胞增殖的影响。流式细胞仪检测细胞周期分布。Western blot法检测粉防己碱处理前后HeLa细胞中Chk1/2蛋白表达情况。结果:粉防己碱对HeLa细胞的增殖抑制作用呈时间浓度相关性并可引起S期阻滞,粉防己碱作用后HeLa细胞Chk1/2蛋白的表达较对照组减少。可能与细胞周期阻滞的分子机制相关。结论:粉防己碱可以抑制宫颈癌HeLa的增殖,其诱导S期阻滞的分子机制可能与Chk1/2蛋白相关。

Objective：To investigate the anticancer activities of tetrandrine on HeLa cells and the molecular mechanisms.Methods： The proliferation inhibitory rates of HeLa cells induced by tetrandrine in different concentrations and time were measured by MTT assays.Cell cycle was analyzed by flow cytometry.The protein expression of Chk1/2 of HeLa cells was detected by Western blot.Results： Tetrandrine inhibited the proliferation of HeLa cells in dosage and time dependent manner.Tetrandrine induced S phase arrest of HeLa cells and the protein expressions of Chk1/2 were decreased in Hela cells treated with Tetrandrine.Conculsion： Tetrandrine can inhibit the proliferation of HeLa cells and has effects on the S phase arrest of HeLa cells.The Chk1/2 proteins maybe associated with the molecular mechanisms of cell cycle arrest.
