摘要
目的:建立GRC—1肾癌细胞凋亡模型并探讨P21WAF1基因对肾癌细胞凋亡的影响。方法:采用顺铂诱导法建立肾癌细胞凋亡模型及转染P21WAF1基因方法观察P21WAF1基因的作用。结果:①用20μmol顺铂诱导GRC—1肾癌细胞。建立了细胞凋亡模型,凋亡细胞占细胞总数为29.4%。②成功转染了P21WAF1基因至GRC—1肾癌细胞;③转染P21WAF1基因的GRC—1肾癌细胞,培养至第10代细胞加入20μmol顺铂后,凋亡细胞占细胞总数为75.6%。④凋亡细胞比率29.4%与75.6%之间存在显著性差异,P<0.01。结论:①成功建立了肾癌细胞凋亡模型;②转染P21WAF1基因对肾癌细胞凋亡有促进作用。
Using Cisplatin inducing renal cancer cells and gene transfection, established the apopto-sis model of renal carcinoma cell and explored P21WAF1 gene's effect on GRC-1 renal carcinoma cell apoptosis. The results showed that: ①Using 20μmol/L cisplatin inducing renal cells, established cell apoptosis model,29. 4% of the total cells were apoptic cells ②successfully transfected P21WAF1 gene into GRC -1 venal cancer ceust ③transfected P21WAF1 cells were cultured to NO. ten generation and were added inti 20μmol/L cilplatin; 75. 6% of the cells were apoptotic cells. ④There is significant difference between ratio of apoptotic cell to total cells 29. 4% AND 75. 6% (P<0. 01). The conclesions were made that renal cancer cell apoptotic model was succfuuy established and P21WAF1 gene improved renal cancer cell apoptosis.
