Relationship between various model in the serum of hepatitis C virus infection and NS 3 antigen of hepatitis C virus in liver tissue and clinical significance

摘要 Objective To probe into the relationship between various model of anti hepatitis C virus, hepatitis C virus RNA in serum and NS 3 antigen of hepatitis C virus in liver tissue and analyse the clinical significance. Methods Forty five patients with hepatitis C related chronic liver disease were studied. When performing liver biopsy, corresponding sera were collected simultaneously. We detected serum HCV RNA using an assay of reverse transcription and nested polymerase chain reaction, serum anti HCV using second generation enzyme linked immunosorbent assay and HCV NS 3 antigen in liver tissue immunohistochemically with murine monoclonal antibodies to NS 3 antigens of hepatitis C virus. Results Positive rates for anti HCV, HCV RNA in serum and for HCV NS 3 antigen in iver tissue increased gradually along with severe degrees and duration of the disease. The positive results of serum anti HCV do not correspond to the positive results for serum HCV RNA in some patients. Some patients were positive for serum anti HCV but were negative for serum HCV RNA. Some patients were positive for serum HCV RNA but were negative for serum anti HCV. The positive rates of the immuno staining for HCV NS3 antigen in liver tissue vearied significantly among the various model for HCV markers in serum. We found the positive rates for HCV NS3 antigen in the liver tissues from serum HCV RNA positive patients were much higher than those from serum anti HCV positive patients. Positive results for HCV NS3 antigen in liver tissue were better corre sponding to positive results for HCV RNA in serum but do not correspond to positive results for anti HCV in serum. Conclusions We analysed the causes of producing above mentioned appearances. Although the positive rates for HCV NS3 antigen detected immunohistochemically in liver tissue were low, the diagnostic tools for hepatitis C are useful. HCV NS3 anti HCV and HCV RNA in serum reflect the condition of patients with HCV infection and the therapeutic effect for patients.
