Objective To find out tenascin expression difference between the HaCaT cells and SCLⅠ, SCL Ⅱ cell lines of skin, we detected the tenascin mRNA expression and elucidated its implication. Methods Primary keratinocytes, spontaneously transformed human skin keratinocytes (HaCaT) and tumor cell lines from squamous cell carcinomas (SCL Ⅰ and SCL Ⅱ) were investigated with Northern blot of Tenascin special gene probe. Rusults Northern hybridization revealed two mRNA species of about 7.8 and 6.2 Kb. Primary keratinocyte and HaCaT cells did not express Tn mRNA but both tumor cell lines did, especislly 7.8 Kb band. Conclusions The specific expression of Tn mRNA in malignant keratinocyte were useful in differentiation between benign and malignant tumors of the skin and may become one of the markers of the turmors.
