单细胞电泳检测放射诱导人淋巴细胞DNA损伤

摘要 单细胞电泳(SCG)具有快速、灵敏和简便等优点,并可检测出单个细胞DNA的损伤程度,这对研究具有高度组织和细胞特异性的肿瘤方面有特殊意义。通过延长SCG技术的标准碱化时间,可以检测出更低剂量放射诱导的人淋巴细胞DNA单链断裂和碱基损伤,使这一技术更加敏感,在低剂量放射作用研究方面有应用价值。 SCG technique is important to detect single-strand breaks and alkalilabile DNA damage in, individual cell. Since, the effects of many reactive intermediates are, tissue and cell-type specific. We modified SCG technique by prolonging the duration of exposure to alkali, it was possible to detect a significant increase in DNA damage of hua man peripheral lymphocytes induced by low doses of Xr-rays. This will make SCG technique useful for investigation concerned with effect of low doses of radiation.
