The effect of progesterone on osteocalcin gene expression was investi gated in order to understand how progesterone acts on steoblast in vitro. For thispurpose, fetal rat calvarial steblasts were cultured in the medium containing10-9 M -10-6 M progesterone respectively fr tw weeks while the cells progressing differentiation. Northern blot analysis showed that progesterne increased the level f stecalcin mRNA in a dse-dependent manner. Among thse doses of progesterne empIoyed in this study, only 10-6 M hormne caused a significant in-duction of steocalcin gene expressin (2. 3 fold, P<0.01 )- Our results demon-strated fr the first time that the ostecalcin gene expression is regulated by progesterone. In additin, the results btained from our study prvides a good evidence for the direct effect of progesterne on steoblast in vitro.
