应用Fura-2/AM和Sulphinpyrazone测定大鼠海马细胞胞浆游离C^(2+)浓度 被引量:2

摘要 本研究进行了Fura-2/AM和Sulphinpyrazone测定大鼠海马细胞胞浆游离Ca^(2+)浓度([Ca^(2+)]i)的方法学探索,发现海马细胞可将Fura-2/AM外排和房室化,质膜有机阴离子转运抑制剂Sulphinpyrazone能阻断这些过程,且对细胞Ca^(2+)应答无不良影响,提高了检测的灵敏度。结论,Sulphinpyrazone可做为海马细胞[Ca^(2+)]_1测定的常规试剂。 We explored the method of determining Cytosolic free Calcium ions ([Ca2+ ]i)with Fura- 2/AM and Sul-phinpyraxone in hippo - Campal cells of rats. It was found that there are high rate excretion into the extracellular medium and sequestraion inside intracellular organdies of the cytoplasmic fura - 2 in hippocampal cells. The drug Sulphinpyrazone, a inhibitor of membrane transport system for organic anions. Can block these processes without any untoward effects on the response of these cells to 50mM KCL. These evidences Suggested that Sulphinpyrazone is a useful drug to assure the accuracy of detecting that [Ca2+ ]i in hippo campal cells with Fura- 2/AM.
