花魔芋球茎休眠和萌发与细胞分裂素关系研究 被引量:5

摘要 用酶联免疫法(ELISA)测定了休眠期间花魔芋球茎中细胞分裂素(CK)的含量。在12℃的贮藏温度下,顶芽中的N6-(2-异戊烯基)腺嘌呤+N6-(2-异戊烯基)腺苷(iP+iPA)和玉米素+玉米素核苷(Z+ZR)的含量在收获后的20d内略有增加,然后有所下降,休眠后期其含量升高3至5倍;周皮和薄壁贮藏组织中的含量较低,在休眠后期也有所增加。20℃下,CK变化加快。6-苄基腺嘌呤(BA)和激动素(KT)处理休眠球茎则促进顶芽的萌发和诱导不定芽的产生。 The contents of cytokinins were determined in konjac corms by enzyme linked immunosorbent assay (ELISA) during dormancy.The contents of N 6 (2 isopentenyl) adenine+N 6 (2 isopentenyl) adenosine (iP+iPA) and zeatin+zeatin riboside (Z+ZR) slightly increased in terminal buds of the corms stored at 12℃ within 20 days,then declined between 20 or 50 and 80 days after harvest and again gradually increased 3 to 5 fold during dormancy release.The contents of cytokinins were low in periderm and parenchyma,but increased during dormancy release.The changes of cytokinins were rapider in corms with the increase of storage temperature from 5℃ to 20℃.Applied 6 benzyladenine (BA) and kinetin (KT) at concentrations of 10mg·L -1 and 50mg·L -1 were found to promote the sprouting of terminal buds and induce the differentiation and formation of adventitious buds.The role of cytokinins on the sprouting and bud growth was discussed.
